Overview

PASylated asparaginase is a long-acting recombinant enzyme therapy being developed for the treatment of acute lymphoblastic leukemia (ALL). It consists of the enzyme asparaginase (specifically crisantaspase) modified using XL-protein's proprietary PASylation technology. PASylation involves the genetic fusion or chemical conjugation of the therapeutic protein with a conformationally disordered polypeptide composed of proline, alanine, and serine (PAS) amino acids. This modification increases the hydrodynamic volume of the protein, thereby extending its plasma half-life and duration of therapeutic effect by reducing renal clearance. The drug works by depleting circulating asparagine, an amino acid that leukemic cells are unable to synthesize themselves, leading to cell death. The program is currently in preclinical development through a collaboration between XL-protein and Jazz Pharmaceuticals.
