Drug intelligence / Profile preview
PASylated ferritin nanocarrier is a genetically engineered drug delivery system developed by XL-protein in collaboration with academic partners (including Sapienza University of Rome, University of Verona, and the National Research Council of Italy). The platform utilizes human heavy-chain ferritin (HFt) fused with a proline-alanine-serine (PAS) or proline-alanine-serine-glutamate (PASE) polypeptide shield (PASylation) to extend blood circulation half-life and reduce off-target uptake. Some constructs (such as HFt-MP-PAS and HFt-MP-PASE) incorporate a matrix metalloproteinase (MMP-2/9)-cleavable linker between the ferritin core and the PAS shield. In the tumor microenvironment, MMPs cleave the shield, unmasking the ferritin nanocage to bind Transferrin Receptor 1 (TfR1/CD71) overexpressed on cancer cells, facilitating receptor-mediated endocytosis. This nanocarrier is designed to encapsulate and deliver cytotoxic payloads, such as doxorubicin, ellipticine, mitoxantrone, or Genz-644282, for the treatment of various cancers.
