The pasylated GLP-1 variant is a long-acting, investigational glucagon-like peptide-1 (GLP-1) receptor agonist being co-developed by Ajinomoto Bio-Pharma Services and XL-protein GmbH for the treatment of metabolic diseases, including type 2 diabetes and obesity. The drug utilizes XL-protein's proprietary PASylation technology, which involves the genetic fusion of the GLP-1 peptide to a conformationally disordered polypeptide chain composed of proline, alanine, and serine residues. This modification increases the molecule's hydrodynamic volume, thereby reducing renal clearance and extending its plasma half-life as a biological alternative to PEGylation. The variant is produced using Ajinomoto's Corynex protein expression system, which allows for high-yield manufacturing. Preclinical studies have suggested the potential for weekly or bi-weekly subcutaneous dosing while maintaining high affinity for the GLP-1 receptor.
