Drug intelligence / Profile preview
PASylated human DNase I (also known as PASylated hyperactive human DNase I) is a preclinical-stage, long-acting recombinant biologic being developed by XL-protein in collaboration with Rentschler Biopharma. The therapeutic agent consists of a hyperactive human deoxyribonuclease I (DNase I) enzyme (specifically containing the N74K mutation) genetically fused at its N-terminus to a 400-residue PAS polypeptide (comprising proline, alanine, and serine residues) using XL-protein's proprietary PASylation technology. This modification significantly increases the enzyme's hydrodynamic volume, retarding renal clearance and extending its plasma half-life, while also boosting its hydrolytic activity toward extracellular DNA. PASylated human DNase I is designed to degrade excess extracellular DNA and neutrophil extracellular traps (NETs) that accumulate in thrombo-inflammatory states, offering potential therapeutic utility in inflammation, autoimmune diseases, and cystic fibrosis.
