Overview

PASylated uricase is a recombinant fusion protein consisting of the enzyme urate oxidase (uricase), typically derived from *Aspergillus flavus*, genetically fused with PAS (proline-alanine-serine) polypeptide sequences. This modification is designed to increase the enzyme's plasma half-life and stability while reducing immunogenicity, serving as a biodegradable alternative to PEGylation technology. The enzyme works by catalyzing the oxidation of uric acid into the more soluble and easily excreted compound allantoin. It is primarily being investigated for the treatment of conditions associated with high uric acid levels, including severe gout, hyperuricemia, chemotherapy-induced hyperuricemia, and tumor lysis syndrome. Among the variants, UOX-PAS100 has demonstrated the most significant improvements in half-life and catalytic efficiency compared to the native enzyme.
