PAT-CM1 is a preclinical-stage, cell-penetrating IgM antibody being developed by Patrys Limited as part of its Deoxymab platform. Derived from the 3E10 lupus autoantibody, PAT-CM1 is a full-sized IgM molecule, which provides it with high valency for binding compared to smaller antibody fragments. It possesses the unique ability to cross the blood-brain barrier and penetrate the nucleus of cancer cells, where it binds to DNA and inhibits the DNA damage repair (DDR) response. By interfering with DDR pathways, PAT-CM1 induces apoptosis in cancer cells, particularly those that already harbor genetic instabilities or are being treated with DNA-damaging agents like radiation or chemotherapy. Its ability to target extracellular DNA released by dying tumor cells also allows it to home in on the tumor microenvironment, making it a promising candidate for the treatment of primary and secondary brain cancers as well as various solid tumors.
