PATAS is a first-in-class, synthetic stapled peptide drug candidate developed to directly target insulin resistance, the root cause of type 2 diabetes and related metabolic disorders. It represents the inaugural member of a new class called "Adipeutics," which are therapeutics specifically designed to restore adipocyte (fat cell) function. Mechanistically, PATAS works by modulating the interaction between Alstrom syndrome protein 1 (ALMS1) and Protein kinase C alpha in adipocytes, thereby restoring glucose uptake and normalizing lipid metabolism. Preclinical studies have shown that PATAS improves whole-body insulin resistance, lowers unhealthy ceramides (implicated in cardiovascular dysfunction), reduces glucose intolerance, liver steatosis (fatty liver), and fibrosis. The drug is being developed by AdipoPharma with plans for clinical trials targeting type 2 diabetes and associated comorbidities such as nonalcoholic steatohepatitis (NASH) and fibrosis[1][2][3][4][5][6][7][8].
