pateclizumab

Pateclizumab is a humanized IgG1 kappa monoclonal antibody that targets lymphotoxin alpha (LTα), a cytokine involved in immune regulation and inflammation. By binding to both soluble LTα3 homotrimers and membrane-bound LTα1β2 heterotrimers, pateclizumab blocks their interaction with cognate receptors (LTβR and TNFR), thereby inhibiting downstream inflammatory signaling pathways. It was developed as an immunomodulator for the treatment of rheumatoid arthritis by Genentech/Roche. Clinical trials demonstrated that while pateclizumab was generally well-tolerated and showed evidence of biological activity, it did not achieve statistically significant improvement in clinical endpoints compared to placebo or adalimumab, leading to discontinuation of its development for rheumatoid arthritis[1][3][4][5][6].
