Overview

Sildenafil is a small molecule drug classified as a phosphodiesterase type 5 (PDE5) inhibitor. It was originally developed by Pfizer and is primarily indicated for the treatment of erectile dysfunction (ED) and pulmonary arterial hypertension (PAH). In ED, sildenafil works by inhibiting PDE5 in the corpus cavernosum of the penis to increase cyclic guanosine monophosphate (cGMP), resulting in smooth muscle relaxation and increased blood flow during sexual stimulation. For PAH, it relaxes blood vessels in the lungs to reduce pulmonary blood pressure and improve exercise capacity. Sildenafil does not cause an erection without sexual stimulation[1][2][3][4][5].
