Overview

Pazopanib is an orally administered small molecule antineoplastic agent used primarily for the treatment of advanced renal cell carcinoma and advanced soft tissue sarcoma in adults who have received prior chemotherapy. It acts as a second-generation, multitargeted tyrosine kinase inhibitor, selectively inhibiting vascular endothelial growth factor receptors (VEGFR-1, VEGFR-2, VEGFR-3), platelet-derived growth factor receptors (PDGFR-alpha and PDGFR-beta), and c-kit. By blocking these targets involved in angiogenesis pathways, pazopanib reduces tumor blood vessel formation necessary for tumor survival and growth. Pazopanib was originally developed by GlaxoSmithKline and is now marketed worldwide by Novartis under the brand name Votrient[1][4][5][6].
