pazopanib + cetuximab is a combination therapy investigated for the treatment of incurable recurrent or metastatic head and neck squamous cell carcinoma (HNSCC). This regimen, notably studied by the Washington University School of Medicine, combines pazopanib—a multi-target tyrosine kinase inhibitor (TKI) administered as an oral suspension—with cetuximab, an intravenously administered monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Pazopanib exerts its effect by inhibiting angiogenesis and tumor cell proliferation through the blockade of VEGFR-1/2/3, PDGFR-α/β, and c-KIT. Cetuximab complements this by inhibiting EGFR-mediated signaling pathways that drive tumor growth. Preliminary clinical data from Phase 1b trials indicated significant antitumor activity, including responses in patients previously resistant to cetuximab or platinum-based chemotherapy, establishing a recommended Phase 2 dose for the combination.
