Overview

Pazopanib + topotecan is a combination of two small molecule drugs used primarily in oncology research and clinical trials. Pazopanib is a second-generation multitargeted tyrosine kinase inhibitor that blocks vascular endothelial growth factor receptor 1 (VEGFR1), vascular endothelial growth factor receptor 2 (VEGFR2), vascular endothelial growth factor receptor 3 (VEGFR3), platelet-derived growth factor receptor alpha (PDGFRA), platelet-derived growth factor receptor beta (PDGFRB), and c-Kit, thereby inhibiting angiogenesis and tumor blood vessel formation[6]. Topotecan is a DNA topoisomerase I inhibitor that induces DNA damage by stabilizing the cleavable complex between DNA and the enzyme, leading to cell death. The combination has shown synergistic anti-tumor activity in preclinical models of gynecologic cancers, sarcomas, triple-negative breast cancer, and anaplastic thyroid cancer by targeting both tumor cells directly (via DNA damage) and the tumor microenvironment (via antiangiogenic effects)[4][5][8]. This regimen has been explored as maintenance therapy or for relapsed disease but with limited objective responses in some clinical settings[1].
