pazopanib + trabectedin + ipilimumab + nivolumab is an experimental combination therapy regimen being evaluated in the TraPIN Phase I clinical trial (NCT04356807) at the M.D. Anderson Cancer Center. This multi-modal approach targets recurrent soft tissue sarcomas in pediatric and young adult patients by combining the anti-angiogenic and anti-proliferative effects of pazopanib (a multi-kinase inhibitor), the DNA-binding cytotoxic activity of trabectedin, and dual immune checkpoint blockade with ipilimumab (anti-CTLA-4) and nivolumab (anti-PD-1). The study aims to determine the safety and recommended Phase II dose of this intensive combination, building upon the SAINT trial's chemotherapy-immunotherapy backbone by adding a targeted tyrosine kinase inhibitor to enhance efficacy in refractory sarcoma populations.
