PB-223 is a chimeric IgG1 monoclonal antibody developed by Precision Biologics through immune engineering and affinity maturation of the earlier mAb NEO-102 (Ensituximab). It specifically targets a unique core 2 O-glycan variant of MUC5AC, which is expressed on certain cancer cells. Preclinical and clinical data indicate that PB-223 binds to and kills colorectal and pancreatic cancer cells via antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Compared to its predecessor NEO-102, PB-223 demonstrates stronger binding to cancer cell lines expressing core 2 O-glycans, covers a broader spectrum of human tumor types while sparing normal tissues, and can internalize into target cancer cells. The drug is being developed for the treatment of solid tumors[1][2][4].
