Overview

PB-MMAE-5 is an investigational antibody-drug conjugate (ADC) developed by Precision Biologics. It consists of the monoclonal antibody PB-223 conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a cysteine-based, cleavable mc-vc-PABc linker. The drug-to-antibody ratio (DAR) for PB-MMAE-5 is approximately 3.92, which falls within the optimal range for ADC efficacy and safety. The ADC targets cancer cells expressing core 2 O-glycans and has demonstrated potent in vitro cytotoxicity against human prostate, triple positive and triple negative breast, lung, colon, and pancreatic cancer cell lines. In vivo studies in NOD-SCID mice with OV-90 xenografts showed significant anti-tumor activity at doses of 1 mg/kg and 3 mg/kg administered weekly for three weeks; higher doses resulted in greater tumor volume reduction compared to controls or lower doses. Toxicology studies in rats indicated that PB-MMAE-5 was well tolerated with no signs of distress or weight loss after administration. Stability testing revealed that the ADC remains stable in human plasma over 14 days[1][2]. MMAE acts as a microtubule inhibitor by blocking tubulin polymerization after intracellular release from the linker[4][7].
