PB-MMAE-6 is an investigational antibody-drug conjugate (ADC) developed by Precision Biologics. It consists of the monoclonal antibody PB-223 conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a cysteine-based, protease-cleavable mc-vc-PABc linker. The drug-to-antibody ratio (DAR) for this ADC is 4.15, indicating a high payload loading per antibody molecule. PB-MMAE-6 targets cancer cells expressing core 2 O-glycans and has demonstrated strong binding affinity and potent cytotoxicity against OV-90 human ovarian cancer cells in vitro, with 80% cell death observed five days post-treatment. While most in vivo efficacy and safety data are available for the closely related clone PB-MMAE-5, all three ADCs (PB-MMAE-2, -5, -6) share similar binding and killing profiles in preclinical models[3]. MMAE acts as a microtubule inhibitor that blocks tubulin polymerization, leading to cell cycle arrest at G2/M phase and subsequent apoptosis[3][5]. The linker ensures stability in circulation but allows release of MMAE inside target cells following endocytosis.
