PB-TL012 is a radioligand therapy (RLT) currently in preclinical development by Primo Biotechnology for the treatment of metastatic colorectal cancer (mCRC). The drug consists of a targeting ligand directed against Folate Hydrolase 1 (FOLH1), also known as Prostate-Specific Membrane Antigen (PSMA), conjugated to the therapeutic beta-emitting radioisotope Lutetium-177 (177Lu). Although FOLH1 is a well-characterized target in prostate cancer, it is also highly expressed in the neovasculature of various other solid tumors, including colorectal cancer. PB-TL012 is designed to bind specifically to FOLH1-expressing cells and deliver localized ionizing radiation, which induces DNA damage and subsequent apoptosis in tumor cells and the supporting tumor microenvironment.
