PB43D is a highly potent, humanized monoclonal antibody of the IgG1 isotype developed as a passive immunotherapy targeting tau protein pathology. It is designed to bind and clear hyperphosphorylated tau, which forms neurofibrillary tangles—a hallmark of Alzheimer’s disease (AD) and other tauopathies such as frontotemporal dementias, progressive supranuclear palsy, cortico-basal degeneration, Pick disease, chronic traumatic encephalopathy, Guam Parkinsonism dementia, and Down syndrome-associated AD. Uniquely among anti-tau antibodies in development or clinical use, PB43D has demonstrated preclinical efficacy in clearing both tau and amyloid-beta (Aβ) pathologies simultaneously and rescuing cognitive impairment in transgenic mouse models of AD. Its mechanism involves inhibiting the seeding and spread of pathological tau aggregates. The antibody has been humanized for clinical development; pre-IND studies are ongoing with plans for Phase 1 trials in AD patients. Developed by Phanes Biotech, PB43D represents a next-generation approach to immunotherapy for neurodegenerative diseases characterized by abnormal protein aggregation[1][2][3][4][5].
