PBA-0091 is a bifunctional fusion protein (BFP) developed by Pure Biologics for the treatment of solid tumors, including head and neck cancer, sarcomas, and triple-negative breast cancer. It is the lead candidate of the PB003G program. The molecule is designed to target the GARP-TGFβ1 complex, which is highly expressed on regulatory T cells (Tregs) and tumor cells, where it mediates immunosuppression. PBA-0091 consists of a monoclonal antibody targeting this complex fused to ULBP2, a natural ligand for the NKG2D receptor. This dual-action approach aims to neutralize the immunosuppressive activity of TGFβ1 while simultaneously recruiting and activating cytotoxic natural killer (NK) and T cells to the tumor microenvironment to eliminate both immunosuppressive Tregs and tumor cells.
