Overview
PBA-0111 is an afucosylated, fully human monoclonal antibody targeting GARP (LRRC32), developed by Pure Biologics for the treatment of solid tumors. Its mechanism involves inhibiting LRRC32 (GARP), a protein that plays a role in immunosuppression within the tumor microenvironment. By blocking and eliminating regulatory T cells, PBA-0111 aims to overcome immunosuppression and enable the immune system to target and destroy tumor cells. The drug is designed to combine immune checkpoint blockade with direct cytotoxic activity against cancer cells. It is currently being evaluated in early-phase clinical trials for indications including soft tissue sarcoma, squamous cell carcinoma of the head and neck, and triple negative breast cancer[2][3][4][6].
