PBA-0405 is an engineered antibody-based immunotherapeutic developed by Pure Biologics for the treatment of solid tumors and selected hematological malignancies. It functions as a bispecific killer engager (BIKE), specifically targeting the receptor tyrosine kinase-like orphan receptor 1 (ROR1) expressed on tumor cells, and has increased binding affinity to the CD16 Fc receptor on natural killer (NK) cells[1][3][4]. By engaging both ROR1 on tumor cells and CD16 on NK cells, PBA-0405 stimulates NK cell-mediated cytotoxicity through antibody-dependent cellular cytotoxicity (ADCC), leading to direct killing of cancer cells[1][4]. The molecule is afucosylated IgG1 class antibody, enhancing its immune-stimulating properties[4]. Indications under investigation include triple-negative breast cancer, head and neck squamous cell carcinoma, soft tissue sarcoma, and B-cell leukemias such as chronic lymphocytic leukemia[2][3][6].
