pBC.1 is a multi-antigen plasmid DNA (pDNA) vaccine designed for the prevention and treatment of breast cancer. Developed through a collaboration involving the National Cancer Institute (NCI), Mayo Clinic, and MRIGlobal, the vaccine utilizes the pUMVC4a vector and 2A peptide technology to enable the coordinated expression of multiple tumor-associated antigens (TAAs). Specifically, pBC.1 encodes the extracellular and intracellular domains of Human Epidermal Growth Factor Receptor 2 (HER2) and Mucin 1 (MUC1). As a gene therapy modality, it offers transient, non-integrating expression of these antigens to stimulate an immune response against breast cancer cells. The vaccine is formulated in saline sodium citrate (SSC) to maintain structural integrity, specifically supercoiled topology, which serves as a critical quality attribute for functional potency.
