Overview

**PBCAR19B** is an allogeneic, immune-evading, anti-CD19 CAR T cell therapy designed for relapsed or refractory B-cell non-Hodgkin lymphoma and other CD19+ malignancies. Developed using gene-editing technologies, PBCAR19B T cells are engineered to enhance persistence and evade host immune rejection. The CAR construct is inserted into the T Cell Receptor Alpha Constant (TRAC) locus to prevent endogenous TCR–mediated graft-versus-host disease (GvHD). Additionally, the product integrates a short hairpin RNA (shRNA) to knock down beta-2 microglobulin, reducing major histocompatibility complex (MHC) class I expression and minimizing cytotoxic T cell-mediated clearance. An HLA-E transgene is included to further reduce natural killer (NK) cell–mediated rejection[1][4][5][7]. This multiplexed engineering strategy aims to improve persistence, safety, and efficacy compared to conventional allogeneic CAR T cell approaches.
