Overview

PBCAR269A is an investigational allogeneic chimeric antigen receptor (CAR) T cell therapy developed by Precision BioSciences for the treatment of relapsed or refractory multiple myeloma. It is manufactured from healthy donor T cells that are genetically modified using Precision's proprietary ARCUS genome editing platform to express a CAR targeting B-cell maturation antigen (BCMA), a protein commonly found on malignant plasma cells in multiple myeloma. As an "off-the-shelf" therapy, PBCAR269A does not require patient-specific cell collection, potentially allowing for more rapid and scalable treatment compared to autologous CAR-T therapies. The drug has been evaluated in Phase 1/2a clinical trials as both monotherapy and in combination with nirogacestat, a gamma secretase inhibitor, to enhance BCMA expression on target cells[1][4][5][6][7][8].
