pBCAR3-phosphopeptide is a synthetic phosphorylated peptide derived from the breast cancer antiestrogen resistance protein 3 (BCAR3), specifically corresponding to amino acids 126-134 of the protein. This phosphopeptide contains a phosphorylated threonine residue and has been developed as a tumor-associated antigen for use in cancer immunotherapy. It is designed to elicit T-cell responses against cancer cells presenting this post-translationally modified epitope via MHC molecules. Preclinical studies demonstrated that pBCAR3-phosphopeptide is immunogenic in mice and humans, with T-cells specific for this peptide capable of controlling tumor growth in xenograft models. In early-phase clinical trials as part of multi-peptide vaccines (often combined with other phosphopeptides such as pIRS2), it was shown to be safe and able to induce immune responses in some patients with melanoma or other cancers[3][4][5][6][7][8]. The mechanism involves presentation by MHC class I molecules and recognition by CD8+ T-cells, targeting tumor cells expressing the phosphorylated form of BCAR3.
