PBD-C06

PBD-C06 is a humanized and deimmunized monoclonal antibody developed by Vivoryon Therapeutics (formerly Probiodrug) for the treatment of Alzheimer's disease. It specifically targets amyloid beta-protein with an N-terminal pyroglutamate modification at position 3 (AβN3pE or pGlu3-Aβ), a form of amyloid beta known to aggregate into neurotoxic oligomers implicated in Alzheimer’s pathology. The antibody was engineered to bind with high specificity and avidity to pGlu3-Aβ monomers, oligomers, and fibrils—including mixed aggregates with unmodified Aβ—while avoiding complement-mediated inflammatory responses by eliminating C1q binding. This design aims to reduce the risk of vasogenic edema and other vascular-related inflammation associated with anti-amyloid therapies. Preclinical studies have shown that PBD-C06 can reduce N3pE amyloid plaque load in animal models without increasing microhemorrhages or inducing significant immune activation[1][4][5][7][9].
