PBENZ-DBRMD is a preclinical-stage small molecule inhibitor targeting Iodothyronine Deiodinase Type 3 (DIO3), developed by Dio Tree. DIO3 is an enzyme responsible for the inactivation of thyroid hormones (T3 and T4) and is frequently upregulated in various malignancies, including high-grade serous ovarian cancer (HGSOC), lung, breast, colon, and pancreatic cancers. Overexpression of DIO3 in the tumor microenvironment is thought to contribute to immune evasion and tumor progression by depleting local thyroid hormone levels. By inhibiting DIO3, PBENZ-DBRMD aims to restore local thyroid hormone signaling, potentially enhancing anti-tumor immune responses and inhibiting cancer cell proliferation.
