Overview

PBF-677 is a potent and selective, orally administered small molecule that acts as an antagonist of the adenosine A3 receptor. Developed by Palobiofarma, it is designed as a first-in-class, gastrointestinal-restricted therapy for inflammatory bowel diseases (IBD), with primary clinical focus on ulcerative colitis. In Phase IIa clinical trials in patients with mild to moderate ulcerative colitis, PBF-677 demonstrated statistically significant improvements in rectal bleeding and stool frequency when used in combination with standard-of-care mesalazine (5-ASA). The drug was found to be safe and well tolerated. Development for glaucoma and irritable bowel syndrome has been discontinued[1][3][5][7][8].
