Overview

PBF-999 is a novel, orally bioavailable small molecule drug that acts as a dual inhibitor of adenosine A2A receptor (A2AR; ADORA2A) and phosphodiesterase 10A (PDE-10A). It was discovered and developed by Palobiofarma. The drug is designed to modulate immune responses and inhibit tumor growth through two main mechanisms. By antagonizing the adenosine A2A receptor on T-lymphocytes, PBF-999 blocks tumor-released adenosine from suppressing T-cell activity, thereby promoting anti-tumor immunity. Simultaneously, inhibition of PDE-10A prevents degradation of cyclic guanosine monophosphate (cGMP), leading to activation of cGMP-dependent signaling pathways that induce apoptosis in tumor cells and may also impact neurological function. PBF-999 has received Orphan Drug Designation from both the FDA and EMA for Prader-Willi syndrome (PWS) and Rare Pediatric Disease Designation from the FDA for this indication. It has also been investigated in Huntington’s disease and advanced solid tumors but development for these indications has been discontinued[1][2][3][4][5][6][7].
