PBI-0110

PBI-0110 is a synthetic small molecule drug candidate originally developed by ProMetic Life Sciences (now Liminal BioSciences). It belongs to a chemical series of substituted phenylalkanoic acid derivatives designed to modulate G-protein coupled receptors (GPCRs) involved in inflammatory and fibrotic pathways. Specifically, PBI-0110 acts as a dual-acting agent, serving as an agonist of G-protein coupled receptor 40 (GPR40, also known as Free Fatty Acid Receptor 1) and an antagonist of G-protein coupled receptor 84 (GPR84). By activating GPR40 and inhibiting GPR84, the compound aims to reduce the activation of macrophages and fibroblasts, thereby inhibiting the production of pro-inflammatory cytokines and the deposition of extracellular matrix. PBI-0110 was investigated for its potential in treating various fibrotic conditions, including idiopathic pulmonary fibrosis (IPF), chronic kidney disease (CKD), and non-alcoholic steatohepatitis (NASH). However, its development was largely deprioritized in favor of the more advanced lead candidate, PBI-4050.
