Overview

PBMCCAR70-NKT is a chimeric antigen receptor (CAR)-engineered invariant natural killer T (iNKT) cell therapy derived from peripheral blood mononuclear cells (PBMCs). It is specifically designed to target CD70, a tumor-associated antigen that is frequently upregulated in acute myeloid leukemia (AML), particularly following treatment with hypomethylating agents (HMAs) such as azacitidine or decitabine. The therapy leverages the unique properties of iNKT cells, which possess both innate and adaptive immune functions, to induce potent cytotoxicity against CD70-expressing leukemic blasts. In preclinical studies, PBMCCAR70-NKT has demonstrated synergistic antitumor activity when combined with HMAs. While effective, it is often compared to hematopoietic stem and progenitor cell (HSPC)-derived versions (AlloCAR70-NKT), which may offer enhanced persistence and off-the-shelf utility.
