pBTE-Fc is a recombinant bispecific T-cell engager (BTE) fusion protein containing an Fc region, designed to target the tumor-associated antigen interleukin-13 receptor alpha 2 (IL13Rα2). It is utilized in a novel immunotherapeutic strategy where naïve T cells are pre-complexed with the BTE (pBTE) prior to intrathecal delivery. This approach aims to treat central nervous system (CNS) malignancies, such as glioblastoma (GBM) and leptomeningeal disease (LMD), by bypassing the blood-tumor and blood-CSF barriers. The Fc fusion construct is designed to improve the retention and stability of the engager on the T-cell surface, enhancing its anti-tumor activity within the immunologically cold tumor microenvironment of the CNS.
