PC-13 is a potential first-in-class small molecule dual inhibitor that concurrently targets phosphoinositide 3-kinase (PI3K) and cyclin-dependent kinases 4 and 6 (CDK4/6). Developed for the treatment of breast cancer, PC-13 is designed to provide a synergistic therapeutic effect by simultaneously blocking two key pathways involved in tumor cell proliferation and survival, which may help overcome resistance associated with single-target inhibition. Preclinical data indicates that PC-13 possesses nanomolar potency against its targets and demonstrates significant anti-tumor activity in T47D xenograft models, showing efficacy comparable to the combination of palbociclib and buparlisib. The compound has shown high selectivity across kinase panels and a promising safety profile in early studies.
