PC-61 is a rat monoclonal antibody (IgG1 isotype) that specifically targets the alpha chain of the interleukin-2 receptor (IL-2Rα), also known as CD25. In murine models, PC-61 is widely utilized as a research tool to deplete CD4+CD25+Foxp3+ regulatory T cells (Tregs), thereby alleviating immune suppression. This depletion is often used to study the role of Tregs in transplant rejection, autoimmune diseases, and tumor immunity. While the original clone is a rat IgG1, engineered versions with optimized Fc domains (e.g., mouse IgG2a) have been developed to enhance intratumoral Treg depletion via antibody-dependent cellular cytotoxicity (ADCC). The antibody works by blocking the binding of IL-2 to its high-affinity receptor and by inducing the clearance of CD25-high cells through effector-mediated mechanisms.
