PCART3 is an autologous chimeric antigen receptor T-cell (CAR-T) therapy being developed by MediSix Therapeutics for the treatment of T-cell malignancies, specifically peripheral T-cell lymphoma (PTCL) and cutaneous T-cell lymphoma (CTCL). The therapy targets CD3, a protein highly and uniformly expressed on malignant T-cells. To overcome the challenge of 'fratricide'—a phenomenon where CAR-T cells eliminate each other because they also express the target antigen—PCART3 utilizes MediSix's proprietary Protein Expression BLocker (PEBL) technology. PEBL prevents the surface expression of the TCR/CD3 complex on the engineered CAR-T cells, allowing them to specifically target and eliminate CD3-positive tumor cells without self-destruction. The therapy is currently in the IND-enabling stage of development.
