Overview

PCBP2 siRNA nanoparticles are an experimental RNA interference (RNAi) therapeutic consisting of small interfering RNA (siRNA) encapsulated within a nanoparticle delivery system. The siRNA is designed to target and silence the poly(rC)-binding protein 2 (PCBP2) gene, also known as αCP2. PCBP2 is an RNA-binding protein that plays a critical role in stabilizing type I collagen mRNA in myofibroblasts and pancreatic stellate cells. By silencing PCBP2, the therapy destabilizes collagen mRNA, leading to a reduction in collagen deposition and the normalization of the dense, fibrotic tumor stroma characteristic of pancreatic ductal adenocarcinoma (PDAC). This stromal normalization is intended to alleviate physical barriers to drug delivery and immune cell infiltration, thereby enhancing the efficacy of other treatments such as anti-PD-L1 immunotherapy. The program is currently in preclinical development, primarily associated with research from the University of Missouri-Kansas City.
