PCDC is an anti-CDCP1 antibody-drug conjugate (ADC) being developed by Chiome Bioscience for the treatment of solid tumors. The drug candidate, also known as h14A043-ATAC, consists of a humanized anti-CDCP1 antibody engineered for low binding to bone marrow hematopoietic stem cells to minimize hematological toxicity. It is conjugated to an amanitin derivative payload with a drug-antibody ratio (DAR) of 2 using site-specific conjugation technology. PCDC targets CUB domain-containing protein 1 (CDCP1), a transmembrane protein highly expressed in various solid cancers including lung, breast, colorectal, pancreatic, prostate, and ovarian cancers. Upon binding to CDCP1, the ADC undergoes rapid internalization and efficient lysosomal trafficking, releasing the cytotoxic payload to induce cell death.
