Overview

pcDNA-p53 is a mammalian expression plasmid construct designed to deliver and overexpress the human TP53 gene, which encodes the p53 tumor suppressor protein. The plasmid typically utilizes a pcDNA3.1 vector backbone containing a Cytomegalovirus (CMV) promoter to drive high-level, constitutive expression of p53 in eukaryotic cells. In the context of oncology research, specifically cervical cancer, pcDNA-p53 is used to restore p53 function in cells where it has been inactivated or degraded, such as by the HPV16 E6 oncoprotein. The resulting increase in p53 levels triggers downstream pathways involved in cell cycle arrest, DNA repair, and apoptosis, thereby inhibiting tumor cell proliferation, migration, and metastasis. In preclinical studies, transfection with pcDNA-p53 has been shown to reverse epithelial-mesenchymal transition (EMT) and reduce lung metastasis in mouse models.
