pcDNA3.1-EN2 is a recombinant DNA plasmid vector designed for the overexpression of the Engrailed Homeobox 2 (EN2) protein in mammalian cells. In the context of oncology research, particularly non-small cell lung cancer (NSCLC), this construct is utilized to investigate the tumor-suppressive role of EN2. Overexpression of EN2 via this plasmid has been shown to inhibit the proliferation, migration, and invasion of lung cancer cells, such as the A549 cell line. Mechanistically, the construct works by regulating the G1/S phase transition and suppressing the expression and nuclear translocation of β-catenin, thereby modulating the Wnt/β-catenin signaling pathway.
