Overview

pCI-E5Multi is an experimental **DNA plasmid vaccine** encoding a synthetic, codon-optimized, non-oncogenic multiepitope version of the **human papillomavirus type 16 E5** antigen. The construct was designed by duplicating two previously described HPV16 E5 cytotoxic T-lymphocyte epitope coding sequences to create a \"harmless\" E5 sequence that preserves immunogenicity while aiming to eliminate oncogenic risk. In preclinical mouse studies using the HPV16-positive C3 tumor model, intramuscular and electroporation-mediated administration of pCI-E5Multi induced E5-specific cellular immune responses, delayed tumor growth, and reduced tumor burden, supporting its use as a therapeutic immunotherapy approach for **HPV16-associated premalignant lesions and cancers**, particularly settings where E5 is still expressed.
