Overview

PCSK9 targeting immunotherapy refers to a therapeutic approach that inhibits proprotein convertase subtilisin/kexin type 9 (PCSK9) to modulate immune responses, particularly in the context of cancer treatment. PCSK9 is best known for its role in cholesterol metabolism, but recent research has shown that inhibiting PCSK9 can enhance antitumor immunity by increasing CD8+ T cell infiltration and reducing regulatory T cell (Treg) populations within the tumor microenvironment. This dual effect not only potentiates the efficacy of immune checkpoint inhibitors such as anti-PD-1/PD-L1 antibodies but also offers a non-cytotoxic alternative with a favorable safety profile compared to traditional chemotherapies. The approach is being explored both as monotherapy and in combination with other immunotherapies, such as anti-CD137 agonists, showing promise for improved outcomes in cancers like non-small cell lung cancer (NSCLC). Administration is typically via subcutaneous injection[2][3][5].
