Overview

The bivalent PCSK9 vaccine is a peptide-based vaccine formulated using virus-like particles (VLPs) to display two distinct epitopes (amino acids 153–163 and 207–223) from the proprotein convertase subtilisin/kexin type 9 (PCSK9) protein[1][6][8]. By eliciting a robust antibody response that targets both epitopes, the vaccine reduces serum PCSK9 levels, increases hepatic LDL receptor (LDL-R) expression, and lowers circulating LDL cholesterol. The vaccine aims to overcome immune tolerance to the self-antigen PCSK9 and function as a long-acting, cost-effective therapy for hypercholesterolemia and atherosclerotic cardiovascular disease. Preclinical studies in mice and nonhuman primates have shown strong immunogenicity, lipid-lowering efficacy, and a favorable safety profile[1][2][6][8].
