PCUR-101

PCUR-101 is an investigational small molecule STAT3 inhibitor developed by Pellficure Pharmaceuticals for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The drug is a synthetic form of plumbagin, a naphthoquinone medicinal agent naturally derived from plants. Its primary mechanism of action involves the inhibition of Signal Transducer and Activator of Transcription 3 (STAT3), a transcription factor that plays a critical role in the progression and treatment resistance of prostate cancer. PCUR-101 was evaluated in Phase I clinical trials both as a monotherapy and in combination with androgen deprivation therapies, including dutasteride or abiraterone/prednisone. While the drug demonstrated a favorable safety profile and biological activity, such as reductions in IL-6 and putrescine levels, its development was discontinued to allow for reformulation aimed at achieving higher dosing capabilities.
