Overview

PCV10 is a 10-valent pneumococcal conjugate vaccine (specifically the brand Synflorix) designed to provide active immunity against 10 serotypes of Streptococcus pneumoniae. In the context of a cluster randomized trial conducted by Nagasaki University in Nha Trang, Vietnam (NCT02961231), the vaccine was evaluated using a simplified "0p+1" dosing schedule. This regimen involves administering a single dose of the vaccine at 12 months of age, omitting the standard primary infant series. The trial aimed to assess whether this reduced-dose schedule could effectively maintain herd immunity and prevent pneumococcal carriage and disease in a population where vaccine-type circulation had already been lowered by a catch-up campaign. The vaccine targets serotypes 1, 4, 5, 6B, 7F, 9V, 14, 18C, 19F, and 23F, which are conjugated to carrier proteins such as non-typeable Haemophilus influenzae protein D, tetanus toxoid, and diphtheria toxoid to enhance the immune response in young children.
