PCZC-SN38 is a first-in-class zip code drug conjugate (ZDC) developed by Kodikaz Therapeutic Solutions for the treatment of pancreatic ductal adenocarcinoma (PDAC). The platform utilizes specific DNA sequences, termed "Zip Codes" (ZC), derived from transposon sequences (specifically the AluSc8 sequence) that naturally facilitate cell-specific genetic transfer via circulating tumor DNA (ctDNA). In PCZC-SN38, the AluSc8 sequence is conjugated to the topoisomerase I inhibitor SN38 (the active metabolite of irinotecan) via a CL2A linker. This approach aims to selectively deliver the cytotoxic payload to pancreatic cancer cells while sparing healthy tissue, thereby overcoming the toxicity and resistance issues associated with systemic chemotherapy like irinotecan. Preclinical studies in patient-derived xenograft (PDX) models have demonstrated significant tumor growth inhibition and improved tolerability compared to irinotecan.
