Overview

PD-0325901 + dacomitinib is an investigational oral combination therapy consisting of two small molecule inhibitors: PD-0325901, a selective MEK1/2 inhibitor, and dacomitinib, an irreversible pan-HER (epidermal growth factor receptor family) inhibitor. This combination was developed to target tumors with KRAS mutations, which often activate the MAPK pathway and are resistant to single-agent MEK inhibition due to feedback activation of HER-family receptors. The rationale for combining these agents is to simultaneously block both the MAPK pathway (via MEK inhibition) and upstream HER-mediated signaling (via pan-HER inhibition), aiming for improved antitumor efficacy in cancers such as non-small cell lung cancer (NSCLC), colorectal cancer, and pancreatic cancer harboring KRAS mutations. Clinical studies have shown preliminary signs of antitumor activity but also significant toxicity that limits long-term use[1][4][7].
