PD-1 + DC-NK cells refers to a combination immunotherapy approach that integrates programmed cell death protein 1 (PD-1) blockade with dendritic cell–natural killer (DC-NK) cell therapy. This strategy leverages two synergistic mechanisms: - **PD-1 blockade** involves the use of antibodies to inhibit the interaction between PD-1 on immune cells and its ligand PD-L1, thereby reversing immune suppression in the tumor microenvironment. While traditionally associated with T-cell activation, recent research demonstrates that NK (natural killer) cells also express PD-1 and can be reinvigorated by anti-PD-1 therapy, leading to enhanced antitumor activity[1][2][5]. - **DC-NK cell therapy** utilizes ex vivo expanded or activated NK cells—often in conjunction with dendritic cells—to boost innate and adaptive antitumor immunity. Dendritic cells can prime both T-cells and NK cells, while cross-talk between these populations is critical for robust immune responses against cancer[4]. The combination aims to overcome resistance seen in some tumors by simultaneously activating both innate (NK) and adaptive (T-cell mediated) immunity. This approach is under investigation primarily for solid tumors resistant to conventional therapies.
