PD-1 antibody + FOLFIRINOX is a combination therapy consisting of a programmed cell death protein 1 (PD-1) monoclonal antibody (such as pembrolizumab or nivolumab) and the multi-agent chemotherapy regimen known as FOLFIRINOX. The PD-1 antibody is an immune checkpoint inhibitor that blocks the interaction between PD-1 on T cells and its ligands, thereby enhancing anti-tumor immune responses. FOLFIRINOX comprises four chemotherapeutic agents—folinic acid (leucovorin), fluorouracil (5-FU), irinotecan, and oxaliplatin—which work synergistically to inhibit DNA synthesis and repair in rapidly dividing cancer cells. This combination has been investigated primarily for advanced pancreatic cancer, including borderline resectable and locally advanced disease, where it has shown improved response rates and survival compared to chemotherapy alone[2][3][4]. The regimen is under clinical investigation in phase II/III trials.
