PD-1 antibody-deactivated autologous T lymphoid cells is an autologous adoptive cell therapy being investigated for the treatment of advanced malignancies, specifically recurrent platinum-resistant or refractory ovarian cancer. The therapeutic process involves the isolation of T lymphoid cells from the patient's peripheral blood, followed by ex vivo expansion. During the manufacturing process, these T cells are incubated with a monoclonal antibody targeting Programmed cell death protein 1 (PD-1). This antibody binds to and blocks (deactivates) the PD-1 receptors on the T cell surface. When the modified cells are re-infused into the patient, the pre-blocked PD-1 receptors are unable to interact with PD-L1 or PD-L2 ligands expressed by tumor cells or other cells in the tumor microenvironment. This prevents the induction of T-cell exhaustion and maintains the cytotoxic activity of the infused cells against the tumor. This approach is being studied by The Third Affiliated Hospital of Chongqing Medical University in Phase 2 clinical trials.
