Overview

PD-1 blocking scFv refers to a class of engineered single-chain variable fragment (scFv) antibodies designed to bind to and inhibit the Programmed Cell Death Protein 1 (PD-1) receptor. PD-1 is an immune checkpoint receptor expressed on T cells that, when engaged by its ligands (PD-L1 or PD-L2), suppresses T-cell activation and promotes immune tolerance. By blocking this interaction, PD-1 scFvs aim to restore anti-tumor immune responses. These fragments are frequently utilized in advanced immunotherapy research, often encoded within CAR-T cells to provide localized checkpoint blockade (secreted scFvs) or incorporated into oncolytic viruses to enhance the tumor microenvironment's immunogenicity. They are primarily research tools and components of complex biologics rather than standalone approved drugs.
