PD-1 blocking scFv refers to a class of engineered single-chain variable fragment (scFv) antibodies designed to bind to and inhibit the Programmed Cell Death Protein 1 (PD-1) receptor. PD-1 is an immune checkpoint receptor expressed on T cells that, when engaged by its ligands (PD-L1 or PD-L2), suppresses T-cell activation and promotes immune tolerance. By blocking this interaction, PD-1 scFvs aim to restore anti-tumor immune responses. These fragments are frequently utilized in advanced immunotherapy research, often encoded within CAR-T cells to provide localized checkpoint blockade (secreted scFvs) or incorporated into oncolytic viruses to enhance the tumor microenvironment's immunogenicity.
